I have a Q45 MAFto install. Once I splice it in does it require a reprogramming? If so, is this a DIY process to calibrate the car to the bigger MAF?

Yes, why would the stock ECU understand a MAF not meant for it? Either a rom flash for the ECU or some sort of piggy back unit will be need to integrate the MAF.
