How to properly do a DIY boost/vacuum leak test?

Ok so I put together a leak tester with the usual rubber coupling, pvc end, and valve stem from a hardware store. My question is what is the best way to run this test? Do I just bolt the tester down on the intake and pressurize it? Or do I need to unhook and plug other piping and/or hook the tester up in a different location?
